Uncertain significance for Short-rib thoracic dysplasia 11 with or without polydactyly — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_052844.4(DYNC2I2):c.433A>G (p.Met145Val), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DYNC2I2 gene (transcript NM_052844.4) at coding-DNA position 433, where A is replaced by G; at the protein level this means replaces methionine at residue 145 with valine — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces methionine with valine at codon 145 of the WDR34 protein (p.Met145Val). The methionine residue is weakly conserved and there is a small physicochemical difference between methionine and valine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with WDR34-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
