This variant lies in the CRPPA gene (transcript NM_001101426.4) at coding-DNA position 362, where A is replaced by G; at the protein level this means replaces glutamic acid at residue 121 with glycine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 121 of the ISPD protein (p.Glu121Gly). This variant is present in population databases (rs757846188, gnomAD 0.1%). This variant has not been reported in the literature in individuals affected with ISPD-related conditions.
