Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000090.4(COL3A1):c.1455+5G>A, citing LabCorp Variant Classification Summary - May 2015: Variant summary: COL3A1 c.1455+5G>A alters a nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Two predict the variant abolishes the canonical 5' splicing donor site and two predict the variant weakens the 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 4e-06 in 251078 control chromosomes. c.1455+5G>A has been observed in individuals from a family affected with vascular Ehlers-Danlos syndrome (Solyst_2022). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. Based on the evidence outlined above, the variant was classified as VUS-possibly pathogenic.
