NM_000335.5(SCN5A):c.2500A>G (p.Asn834Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N834D variant (also known as c.2500A>G), located in coding exon 15 of the SCN5A gene, results from an A to G substitution at nucleotide position 2500. The asparagine at codon 834 is replaced by aspartic acid, an amino acid with highly similar properties. This alteration was identified in one individual in a long QT genetic testing cohort (Lieve KV et al. Genet Test Mol Biomarkers, 2013 Jul;17:553-61). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
