NM_001103.4(ACTN2):c.721G>A (p.Asp241Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D241N variant (also known as c.721G>A), located in coding exon 8 of the ACTN2 gene, results from a G to A substitution at nucleotide position 721. The aspartic acid at codon 241 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
