But on same aminoacid position the pathogenic mutations p.(Arg270Cys), p.(Arg270Gly) and p.(Arg270Leu) for phenotype waardenburg syndrome are known. The position Arg270 is a hotspot in the homeodomain of the PAX3 gene (Pingault et al. 2010). Multiple lines of computational evidence support a deleterious effect (AGVGD, SIFT, MutationTaster2021, PolyPhen-2).
