Likely pathogenic for Metaphyseal chondrodysplasia, McKusick type — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NC_000009.12:g.35658022TCCTCAGCTTCACAGA[3], citing LabCorp Variant Classification Summary - May 2015: Variant summary: RMRP n.-19_-4[3] is located in the untranscribed region upstream of the RMRP gene region, which is located between the TATA box (-33 to -25) and the transcription initiation site. Other insertions, duplications and triplications in the promoter region of RMRP have been classified as pathogenic (internally and in ClinVar). The frequency of this variant in the general population could not be determined as the technology used for large population databases (ExAC, gnomAD, ESP, 1000G) cannot detect variants of this type. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of n.-19_-4[3] in individuals affected with Cartilage-Hair Hypoplasia and no experimental evidence demonstrating its impact on protein function have been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014. Based on the evidence outlined above, the variant was classified as likely pathogenic.
