This variant lies in the BRAF gene (transcript NM_004333.6) at coding-DNA position 82, where G is replaced by A; at the protein level this means replaces glycine at residue 28 with serine — a missense variant. Submitter rationale: Variant summary: BRAF c.82G>A (p.Gly28Ser) results in a non-conservative amino acid change in the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 6.7e-06 in 149822 control chromosomes (gnomAD v3.1, genomes dataset). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance.
