This variant lies in the FLNA gene (transcript NM_001110556.2) at coding-DNA position 1117, where T is replaced by A; at the protein level this means replaces tyrosine at residue 373 with asparagine — a missense variant. Submitter rationale: Variant summary: FLNA c.1117T>A (p.Tyr373Asn) results in a non-conservative amino acid change located in the Filamin/ABP280 repeat (IPR001298) of the encoded protein sequence. Three of five in-silico tools predict a benign effect of the variant on protein function. The variant was absent in 181309 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1117T>A in individuals affected with Periventricular Nodular Heterotopia 1 and no experimental evidence demonstrating its impact on protein function have been reported.
