This variant lies in the DHCR7 gene (transcript NM_001360.3) at coding-DNA position 1088, where G is replaced by T; at the protein level this means replaces arginine at residue 363 with leucine — a missense variant. Submitter rationale: Variant summary: DHCR7 c.1088G>T (p.Arg363Leu) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 248778 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1088G>T in individuals affected with Smith-Lemli-Opitz Syndrome and no experimental evidence demonstrating its impact on protein function have been reported.
