NM_000360.4(TH):c.1061C>T (p.Ala354Val) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: TH c.1154C>T (p.Ala385Val) results in a non-conservative amino acid change located in the Aromatic amino acid hydroxylase, C-terminal (IPR019774) of the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 1.8e-05 in 164392 control chromosomes (gnomAD). c.1154C>T has been reported in the literature in compound heterozygous individuals affected with tyrosine hydroxylase deficiency (examples: Mak_2010 and Yeung_2011). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. No clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014.
