NM_001042492.3(NF1):c.6206T>G (p.Leu2069Ter) was classified as Pathogenic for Large cafe-au-lait macules with irregular margins; Neurofibroma; Neurofibromatosis, type 1 by Department of Dermatology, Xinhua Hospital, Shanghai Jiao Tong University of Medicine: The c.6143T>G variant in NF1 was absent from population variation database, leading to a premature termination codon and are prone to nonsense-mediated mRNA decay. In addition, the nucleotide is predicted to be high conserved by phastCons and PhyloP. Patient’s phenotype and family history is highly specific for neurofibromatosis type 1 with a single genetic etiology. In summary, the variant meets our criteria to be calssified as pathogenic (PVS1+PM2+PP3+PP4) accroding to standards and guidelines for the interpretation of sequence variants of American College of Medical Genetics and Genomics (ACMG)(Richards et al., 2015).
