NM_001369.3(DNAH5):c.13837G>A (p.Val4613Met) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V4613M variant (also known as c.13837G>A), located in coding exon 79 of the DNAH5 gene, results from a G to A substitution at nucleotide position 13837. The valine at codon 4613 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
