Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_021167.5(GATAD1):c.537C>G (p.Cys179Trp), citing Ambry Variant Classification Scheme 2023: The p.C179W variant (also known as c.537C>G), located in coding exon 4 of the GATAD1 gene, results from a C to G substitution at nucleotide position 537. The cysteine at codon 179 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
