Pathogenic for Autosomal dominant hypohidrotic ectodermal dysplasia syndrome — the classification assigned by Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine to NM_022336.3(EDAR):c.(?_998)_(1347_?)del, citing LMM Criteria. This is a large deletion in the EDAR gene (transcript NM_022336.3) whose exact breakpoints are not precisely mapped. Submitter rationale: The deletion of exons 11 and 12 of the EDAR gene has not been identified in any individual with hypohidrotic ectodermal dysplasia (HED), and was absent from lar ge population studies. Other deletions or loss of function variants have been re ported in individuals with HED. This alteration is then predicted to lead to a t runcated or absent protein. In summary, this variant meets our criteria to be cl assified as pathogenic (http://pcpgm.partners.org/LMM).
