NM_178452.4(DNAAF1):c.(?_-181)_(*77_?)del was classified as Pathogenic for Primary ciliary dyskinesia by Laboratory for Molecular Medicine, Mass General Brigham Personalized Medicine, citing LMM Criteria. This is a large deletion in the DNAAF1 gene (transcript NM_178452.4) whose exact breakpoints are not precisely mapped. Submitter rationale: The (?_-181)_(*77_?)del copy number variant leads to a deletion of the whole DNA AF1 gene. Copy number losses that encompass the DNAAF1 gene and several loss-of- function mutations in this gene in homozygous or compound heterozygous state hav e been reported in 9 individuals with primary ciliary dyskinesia (PCD; Loges 200 9 and Duquesnoy 2009). Three of these individuals had left-right body asymmetry, 3 of them had lobectomy, and at least 6 of them had severe airway diseases incl uding bronchitis, sinusitis, otitis, and bronchiectasis (Loges 2009 and Duquesno y 2009). Copy number losses in the DNAAF1 gene have not been reported in healthy individuals in the DGV database (http://dgv.tcag.ca/dgv/app/home). In summary, this variant meets our criteria to be classified as pathogenic (http://pcpgm.par tners.org/LMM).
