This variant lies in the CPLANE1 gene (transcript NM_001384732.1) at coding-DNA position 8410, where C is replaced by T; at the protein level this means replaces proline at residue 2804 with serine — a missense variant. Submitter rationale: Variant summary: CPLANE1 c.8248C>T (p.Pro2750Ser) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 0.00013 in 250244 control chromosomes. This frequency is not significantly higher than expected for a pathogenic variant in CPLANE1 causing Joubert Syndrome And Related Disorders (0.00013 vs 0.0015), allowing no conclusion about variant significance. c.8248C>T has been reported in the literature in one individual affected with Joubert Syndrome. This report does not provide unequivocal conclusions about association of the variant with Joubert Syndrome And Related Disorders. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
