Uncertain Significance for Catecholaminergic polymorphic ventricular tachycardia — the classification assigned by All of Us Research Program, National Institutes of Health to NM_001035.3(RYR2):c.10495-16AT[2], citing ACMG Guidelines, 2015: This variant causes deletion of two nucleotides in intron 72 of the RYR2 gene. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with RYR2-related disorders in the literature. This variant has been identified in 1/241364 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.

This study involves interpretation of variants in research participants for the purpose of population health screening. Participant phenotype was not available at the time of variant classification. Additional details can be found in publication PMID: 35346344, PMCID: PMC8962531
