Submitter rationale: CFB p.Lys323Glu (c.967A>G) is a missense variant that changes the amino acid at residue 323 from Lysine to Glutamic acid. This variant has been observed in at least one proband affected with atypical hemolytic-uremic syndrome (PMID:35372954;26826462;17182750;29670616). The variant was found to segregate with disease in at least one affected family (PMID:29670616). A de novo occurrence of this variant has been observed in at least one affected individual (PMID:17182750). At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:17182750). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify CFB p.Lys323Glu (c.967A>G) as a pathogenic variant.
