Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_019842.4(KCNQ5):c.1358C>T (p.Thr453Ile), citing LabCorp Variant Classification Summary - May 2015: Variant summary: KCNQ5 c.1415C>T (p.Thr472Ile) results in a non-conservative amino acid change located in the potassium channel, voltage dependent, KCNQ, C-terminal domain (IPR013821) of the encoded protein sequence. Three of four in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251418 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.1415C>T in individuals affected with Intellectual Disability, Autosomal Dominant 46 and no experimental evidence demonstrating its impact on protein function have been reported.
