Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000516.7(GNAS):c.493C>T (p.Arg165Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with cysteine, which is neutral and slightly polar, at codon 165 of the GNAS protein (p.Arg165Cys). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of GNAS-related conditions (PMID: 8388883, 23281139, 25802881). In at least one individual the variant was observed to be de novo. This variant is also known as c.496C>T p.(Arg166Cys). ClinVar contains an entry for this variant (Variation ID: 15932). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt GNAS protein function with a positive predictive value of 95%. Studies have shown that this missense change alters GNAS gene expression (PMID: 8388883). For these reasons, this variant has been classified as Pathogenic.
