Pathogenic for Bilateral frontoparietal polymicrogyria — the classification assigned by Kasturba Medical College, Manipal, Kasturba Medical College, Manipal, Manipal Academy of Higher Education, Manipal, India to NM_201525.4(ADGRG1):c.1408C>T (p.Arg470Ter), citing ACMG Guidelines, 2015. This variant lies in the ADGRG1 gene (transcript NM_201525.4) at coding-DNA position 1408, where C is replaced by T; at the protein level this means converts the codon for arginine at residue 470 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: The stopgain variant, c.1408C>T was observed in heterozygous state in the proband (Desai et al., 2015; Sawal et al., 2018; ClinVar Accession: VCV000158618.19). On Sanger validation and segregation analysis, this variant was found to be in heterozygous state in the proband and her mother but absent in her father. This variant is present in 32 individuals in heterozygous state and absent in homozygous state in gnomAD (v4.1.0) population database. This same variant is absent in heterozygous and/or homozygous state in and our in-house database of 3871 individuals. This variant is predicted to introduce a premature termination codon in the transcript which may either lead to nonsense-mediated mRNA decay of the transcript or the formation of a truncated protein product.
