The NHLBI ESP Exome Variant Project has not identified Gly25Arg in approximately 6,500 individuals of European or African American ethnicity, indicating that it is not a common benign variant in these populations. This amino acid substitution is non-conservative, as an uncharged, non-polar Glycine residue is replaced by a positively charged Arginine residue. Gly25Arg alters a highly conserved position in the ATP-binding domain of the protein, and multiple in silico algorithms predict it is damaging to protein structure/function. Additionally, the variant has been observed as a mosaic and de novo with validated parentage. The variant is found in EPILEPSY panel(s).
