Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251276 control chromosomes. c.347C>A has been observed in the heterozygous state in multiple individuals affected with Beta Thalassemia and segregated with disease in at least two families consistent with autosomal dominant inheritance (Ohga_2003, Da_2021, Parikh_2005, Divoky_1993). These data indicate that the variant is very likely to be associated with disease. This variant is also known as Hb Hradec Kralove (or Hb HK).
