Uncertain significance — the classification assigned by Quest Diagnostics Nichols Institute San Juan Capistrano to NM_000518.4(HBB):c.157G>C (p.Asp53His), citing Quest Diagnostics criteria: The HBB c.157G>C (p.Asp53His) variant (also known as Hb Summer Hill) has been reported to have normal stability, with no significant change in oxygen affinity/cooperativity (PMIDs: 6629828 (1983), 7190137 (1980)). Individuals who are heterozygous for this variant have a normal clinical presentation (HbVar (http://globin.cse.psu.edu/cgi-bin/hbvar/counter), PMID: 6629828 (1983)). This variant has not been reported in large, multi-ethnic general populations (Genome Aggregation Database, http://gnomad.broadinstitute.org). Analysis of this variant using bioinformatics tools for the prediction of the effect of amino acid changes on protein structure and function yielded predictions that this variant is damaging. Based on the available information, we are unable to determine the clinical significance of this variant.
