Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000518.5(HBB):c.22G>A (p.Glu8Lys), citing LabCorp Variant Classification Summary - May 2015: Variant summary: HBB c.22G>A (p.Glu8Lys), also known as Hb G-Siriraj or [beta7(A4)Glu>Lys] in legacy nomenclature, results in a conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change. The variant was absent in 251184 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.22G>A has been observed in the heterozygous and presumed compound heterozygous state in numerous individuals who were either asymptomatic or had very mild features of beta thalassemia minor which were often attributable to other genetic variations in HBB or HBA (example, Chen_2023, Bao_2023, Guan_2022, Li_2022, Lin_2013, Chang_1999, Xu_2020, Blackwell_1972, Rhoda_1986, Tuchinda_965, Ousguine_2025). These report(s) do not provide unequivocal conclusions about association of the variant with Hemoglobinopathy. To our knowledge, no experimental evidence demonstrating an impact on protein function in vitro has been reported.
