Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000518.5(HBB):c.272A>G (p.Glu91Gly), citing LabCorp Variant Classification Summary - May 2015: Variant summary: HBB c.272A>G (p.Glu91Gly), also mentioned as Hb Roseau-Pointe a Pitre, results in a non-conservative amino acid change in the encoded protein sequence. Algorithms developed to predict the effect of missense changes on protein structure and function all suggest that this variant is likely to be disruptive. The variant was absent in 251422 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.272A>G has been observed in at-least one neonate with severe microcytic anemia (Merault_1985). These data do not allow any conclusion about variant significance. A different variant affecting the same codon has been classified as pathogenic by our lab (c.273G>C, p.Glu91Asp), supporting the critical relevance of codon 91 to HBB protein function. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
