Uncertain significance for Glycogen storage disease, type IV; Glycogen storage disease IV, classic hepatic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000158.4(GBE1):c.841A>T (p.Ile281Phe), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine with phenylalanine at codon 281 of the GBE1 protein (p.Ile281Phe). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and phenylalanine. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with GBE1-related conditions. ClinVar contains an entry for this variant (Variation ID: 1525840). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed for this missense variant. However, the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on GBE1 protein function.
