Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.6154C>G (p.Gln2052Glu), citing Ambry Variant Classification Scheme 2023: The p.Q2052E variant (also known as c.6154C>G), located in coding exon 45 of the POLE gene, results from a C to G substitution at nucleotide position 6154. The glutamine at codon 2052 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
