NM_000051.4(ATM):c.300G>C (p.Leu100Phe) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L100F variant (also known as c.300G>C), located in coding exon 3 of the ATM gene, results from a G to C substitution at nucleotide position 300. The leucine at codon 100 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
