Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt SLC12A6 protein function. This variant has not been reported in the literature in individuals affected with SLC12A6-related conditions. This variant is present in population databases (rs770783522, ExAC 0.002%). This sequence change replaces glutamic acid with glycine at codon 5 of the SLC12A6 protein (p.Glu5Gly). The glutamic acid residue is weakly conserved and there is a moderate physicochemical difference between glutamic acid and glycine.
