NM_001382391.1(CSPP1):c.2776C>T (p.Arg926Cys) was classified as Uncertain significance for Joubert syndrome 21 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with cysteine at codon 921 of the CSPP1 protein (p.Arg921Cys). The arginine residue is moderately conserved and there is a large physicochemical difference between arginine and cysteine. This variant is present in population databases (rs373151997, ExAC 0.04%). This variant has not been reported in the literature in individuals affected with CSPP1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
