Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.4532G>A (p.Gly1511Asp), citing Ambry Variant Classification Scheme 2023: The p.G1511D variant (also known as c.4532G>A), located in coding exon 34 of the TSC2 gene, results from a G to A substitution at nucleotide position 4532. The glycine at codon 1511 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
