Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001613.4(ACTA2):c.784A>G (p.Thr262Ala), citing Ambry Variant Classification Scheme 2023: The p.T262A variant (also known as c.784A>G), located in coding exon 6 of the ACTA2 gene, results from an A to G substitution at nucleotide position 784. The threonine at codon 262 is replaced by alanine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
