Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015909.4(NBAS):c.3838C>T (p.Arg1280Trp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine, which is basic and polar, with tryptophan, which is neutral and slightly polar, at codon 1280 of the NBAS protein (p.Arg1280Trp). This variant is present in population databases (rs368280415, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with NBAS-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site.
