Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_000180.4(GUCY2D):c.721+5G>T, citing LabCorp Variant Classification Summary - May 2015: Variant summary: GUCY2D c.721+5G>T alters a conserved nucleotide located close to a canonical splice site and therefore could affect mRNA splicing, leading to a significantly altered protein sequence. Several computational tools predict a significant impact on normal splicing: Three predict the variant weakens a canonical 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant was absent in 228496 control chromosomes. c.721+5G>T has been observed in individual(s) affected with Leber congenital amaurosis (example: Bouzia_2020, and Preising_2007). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
