NM_057175.5(NAA15):c.2591A>G (p.Asn864Ser) was classified as Uncertain significance by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 864 of the NAA15 protein (p.Asn864Ser). This variant is present in population databases (rs201822171, gnomAD 0.06%), and has an allele count higher than expected for a pathogenic variant. This missense change has been observed in individual(s) with autism spectrum disorder (PMID: 37130971). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 1522897). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
