NM_001365951.3(KIF1B):c.3153G>A (p.Met1051Ile) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M1005I variant (also known as c.3015G>A), located in coding exon 27 of the KIF1B gene, results from a G to A substitution at nucleotide position 3015. The methionine at codon 1005 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
