NM_020778.5(ALPK3):c.304G>A (p.Gly102Arg) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G304R variant (also known as c.910G>A), located in coding exon 3 of the ALPK3 gene, results from a G to A substitution at nucleotide position 910. The amino acid change results in glycine to arginine at codon 304, an amino acid with dissimilar properties. However, this change occurs in the last base pair of coding exon 3, which makes it likely to have some effect on normal mRNA splicing. This nucleotide position is highly conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will not have any significant effect on splicing. This amino acid position is highly conserved in available vertebrate species. In addition, as a missense substitution this is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
