Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt INF2 protein function. This variant has not been reported in the literature in individuals affected with INF2-related conditions. This variant is present in population databases (rs778308700, ExAC 0.002%). This sequence change replaces arginine with tryptophan at codon 917 of the INF2 protein (p.Arg917Trp). The arginine residue is moderately conserved and there is a moderate physicochemical difference between arginine and tryptophan.
