Uncertain significance for Genitopatellar syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_012330.4(KAT6B):c.5899A>G (p.Met1967Val), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with valine at codon 1967 of the KAT6B protein (p.Met1967Val). The methionine residue is weakly conserved and there is a small physicochemical difference between methionine and valine. This variant is present in population databases (rs750955934, ExAC 0.006%). This variant has not been reported in the literature in individuals affected with KAT6B-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
