Uncertain significance for Herpes simplex encephalitis, susceptibility to, 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_030930.4(UNC93B1):c.1099G>A (p.Val367Met), citing Invitae Variant Classification Sherloc (09022015): Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. This sequence change replaces valine with methionine at codon 367 of the UNC93B1 protein (p.Val367Met). The valine residue is highly conserved and there is a small physicochemical difference between valine and methionine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate insufficient coverage at this position in the ExAC database. This variant has not been reported in the literature in individuals with UNC93B1-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
