Uncertain significance for Familial hemophagocytic lymphohistiocytosis 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001083116.3(PRF1):c.622A>T (p.Thr208Ser), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the PRF1 gene (transcript NM_001083116.3) at coding-DNA position 622, where A is replaced by T; at the protein level this means replaces threonine at residue 208 with serine — a missense variant. Submitter rationale: This sequence change replaces threonine with serine at codon 208 of the PRF1 protein (p.Thr208Ser). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and serine. This variant is present in population databases (rs769116034, ExAC 0.02%). This variant has not been reported in the literature in individuals affected with PRF1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function output the following: SIFT: "Tolerated"; PolyPhen-2: "Benign"; Align-GVGD: "Class C0". The serine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
