Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_016138.5(COQ7):c.178G>A (p.Glu60Lys), citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with COQ7-related conditions. This variant is present in population databases (rs201819287, gnomAD 0.0009%). This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 60 of the COQ7 protein (p.Glu60Lys).
