Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_172240.3(POC1B):c.517A>G (p.Thr173Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with alanine at codon 173 of the POC1B protein (p.Thr173Ala). The threonine residue is moderately conserved and there is a small physicochemical difference between threonine and alanine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt POC1B protein function. This variant has not been reported in the literature in individuals with POC1B-related conditions. This variant is not present in population databases (ExAC no frequency).
