Pathogenic for Hereditary breast ovarian cancer syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000059.4(BRCA2):c.8488-1G>C, citing Invitae Variant Classification Sherloc (09022015): This sequence change affects an acceptor splice site in intron 19 of the BRCA2 gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with breast and/or ovarian cancer and clinical features of Fanconi Anemia (PMID: 12065746, 22632462, 24607278, 28111427, 29446198, 29790872, 29907814, 30078507, 30103829, 30606148). ClinVar contains an entry for this variant (Variation ID: 1520790). Studies have shown that disruption of this splice site alters mRNA splicing and is expected to lead to the loss of protein expression (PMID: 22632462, 24607278, 25382762).
