Uncertain significance for Osteogenesis imperfecta type 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_022356.4(P3H1):c.1237A>G (p.Thr413Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces threonine with alanine at codon 413 of the P3H1 protein (p.Thr413Ala). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and alanine. The frequency data for this variant in the population databases is considered unreliable, as metrics indicate poor data quality at this position in the ExAC database. This variant has not been reported in the literature in individuals affected with P3H1-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
