NM_020975.6(RET):c.2998G>A (p.Asp1000Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D1000N variant (also known as c.2998G>A), located in coding exon 18 of the RET gene, results from a G to A substitution at nucleotide position 2998. The aspartic acid at codon 1000 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
