Uncertain significance for Developmental and epileptic encephalopathy, 2; Angelman syndrome-like — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NC_000023.11:g.18650485T>A, citing Invitae Variant Classification Sherloc (09022015): Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt CDKL5 protein function. This variant has not been reported in the literature in individuals affected with CDKL5-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant is not present in population databases (ExAC no frequency). This sequence change replaces isoleucine with asparagine at codon 958 of the CDKL5 protein (p.Ile958Asn). The isoleucine residue is weakly conserved and there is a large physicochemical difference between isoleucine and asparagine.
