Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004100.5(EYA4):c.407A>G (p.His136Arg), citing Ambry Variant Classification Scheme 2023: The p.H136R variant (also known as c.407A>G), located in coding exon 6 of the EYA4 gene, results from an A to G substitution at nucleotide position 407. The histidine at codon 136 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
